3.6.7 Aliquoting Description and Scheme - 8:00 AM

8:00 AM Aliquot Assignments: * indicates samples for blind duplicates. The blind duplicate tube is collected after the regular tubes are filled. It would be the fifth tube filled at the 8:00 AM draw. The following blind duplicate tube types will be drawn:

  • 5 mL EDTA (B-D 366452)
  • 5 mL Serum (B-D 366534)
  • 5 mL Citrate (B-D 366415)
  • 5 mL SCAT-1

  • Citrate – After centrifugation, 0.5 mL of plasma is pipetted into cryovials #17–20. These will be blue-capped cryovials. The packed cells remaining in the tube can be added to tube #33. Tube #33 is a red-capped 10.0 mL tube.
  • EDTA – After centrifugation, plasma will be pipetted into cryovials # 21–24. 1.0 mL of plasma is added to clear-capped vial #21. 1.0 mL of plasma is added to white-capped vials #22-24. The packed cells remaining in the tube are added to tube #33.
  • SCAT-1 – After centrifugation, plasma will be pipetted into cryovials #25–28. These will be yellow-capped cryovials. The packed cells remaining in the tube are added to tube #33.
  • Packed Cells – The packed cell tube #33 is a red-capped cryovial that will be frozen and shipped to the LCBR for storage.

Note: Packed cells are only collected from the 8:00 AM draw and not the PM draw. Although the tube holds 10 mL, only 9 mL of material can be added to the tube due to expansion during freezing. The cells from the EDTA and SCAT-1 tubes should be added to tube #33 first and, if there is sufficient space, cells from the citrate tube can also be added for a total volume of 9 mL

  • Serum – 1.0 mL of the serum is pipetted into cryovials #29–32. Tube #29 is a clear-capped cryovial and tubes #30–32 are red-capped cryovials.
  • Blind Duplicates* – The tubes are handled in the same way as the regular collection tubes. The EDTA, Citrate and SCAT-1 tubes are placed on the mixer for approximately 30 seconds, then placed in ice, and centrifuged within 15 to 30 minutes. The Serum tube remains at room temperature for a minimum of 40 minutes to clot before centrifuging.

Once the samples are aliquoted, the EDTA, Citrate and SCAT-1 blood collection tubes can be discarded in the biohazard waste (puncture-proof containers).
